Alteration of the specificity of antitumor antisera by the use of passively administered antibody.

چکیده
Antispleen antibody was produced in burros by immunization with C57BL/6 spleen lymphocytes. The effect of passive administration of this antiserum in inhibiting antibody production to antigenic determinants common to both C57BL/6 lymphocytes and EL4 tumor cells was studied. Simultaneous immunization with EL4 cells and antispleen antibody resulted in specific alteration of anti-EL4 activity so that cross-reactivity to splenic lymphocytes was diminished. These results demonstrate the potential usefulness of passive antibody administration in altering host immunological responsiveness to antigenic determinants on tumor cells.

Advantages of Placenta as a Source for Mass Production of HLA-Antisera
Human Leukocyte Antigens (HLA) of fetus are responsible for induction of HLA antibodies in mother. Exposure to the mismatched paternal HLA antigens in the fetus gives rise to a polyclonal antibody response against paternal HLA antigens. HLA antibodies are present in the maternal circulation of the placenta and are absorbed onto the placenta [1]. The major source of HLA antisera is from sera of ...
متن کاملProduction and Evaluation of Polyclonal Rabbit Anti-Human p53 Antibody Using Bacterially Expressed Glutathione S-transferase-p53 fusion protein
p53 is a key tumor suppressor gene that is targeted for inactivation during human tumorigenesis. In this study, we produced and characterized polyclonal antihuman p53 antibody. The cDNA encoding the completehuman p53 protein was cloned into pGEX-4T-1 and expressed in Escherichia coli as a fusion protein with Schistosoma japonicum glutathione S-transferase (GST). The rabbits were immunized...
متن کاملImmunological response of the rabbit to Vi antigen.
Vi antibody response of rabbits varied depending on whether Vi antigen was administered in particulate or soluble state. Vi antigen in particulate form induced hemagglutinins, bacterial agglutinins, and passive cutaneous anaphylaxis (PCA) antibodies, whereas soluble Vi antigen induced only hemagglutinins. Guinea pigs passively sensitized with antisera against particulate Vi antigen gave PCA rea...
متن کاملRAPID IDENTIFICATION OF Vibrio cholera-01 BY COAGGLUTINATION TEST USING MONO-SPECIFIC ANTIBODY
Mono-specific antisera against Vibrio cholera Ogawa NIH-43 and Vibrio cholera Inaba 35-A3 were prepared from rabbit hyperimmune sera by absorbing against a heterologous strain. Using ammonium sulphate precipitation procedure, gamma globulins were purified and concentrated. To visualize antigen-antibody reaction, gamma globulins were conjugated to Staphylococcus aureus cowan-l (NCTC: 8325) ...
متن کاملThe formalinization of antibody to tumor cells in altering the immune response.
Formalin fixation of passive antibody to tumor cells was effective in modifying subsequently harvested antitumor serum so that the antiserum had diminished reactivity to determinants against which the passively administered antibody was directed. The process of formalin fixation of passive antibody to cells made antigen-antibody bonds more avid without producing nonspecific adherence of antibod...
